Melanotan II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte stimulating hormone (α-MSH), developed at the University of Arizona in the 1990s. It acts as a non-selective agonist at multiple melanocortin receptors including MC1R, MC3R, MC4R, and MC5R.
The peptide structure (Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH₂) includes a lactam bridge between aspartic acid and lysine residues, creating the cyclic configuration that provides enhanced resistance to enzymatic degradation compared to linear peptides.
